Expression and Characterization of A Chitinolytic Enzyme in Chlorella vulgaris

Abstract
The present investigation describes the characterization of a chitinolytic enzyme that catalyzes the degradation of chitin. The kinetics of a novel chitinolytic enzyme designed as Chi-cv expressed from Chlorella vulgaris known as one of typical photosynthetic green alga was partially characterized. Interestingly, Chi-cv activity was observed from the culture supernatant of C. vulgaris cultured in the presence of colloidal chitin. The molecular mass of Chi-cv expressed from C. vulgaris in the presence of colloidal chitin was estimated to be 65 kDa by SDS-PAGE. The optimal pH and temperature of chitinase activity toward the colloidal chitin used as an artificial substrate was pH 7.5 and 30o C, respectively. Especially, chitinase activity has significantly increased by addition of 10 mM NaCl or KCl and 1 mM EDTA in the reaction mixture. The Km and Vmax values toward pNP-(GlcNAC)2 were 0.22 mM and 1.75 nmol/ug/min, respectively. These results suggest that chitinase of C. vulgaris may play a critical role in chitin catabolism when they are heterotrophically adapted from autophototropic condition. Taken together our results demonstrate the existence of chitinolytic enzyme in C. vulgaris for the very first time in the literature.
